Thesis Defense

FIONA PERIS, TecnATox member, defended her PhD last Friday, January 15th at 12:00 p.m in Sala de Graus, School of Medicine and Health Sciences (FMCS). She obtained the highest qualification excellent cum laude.

Title: "ApoE phenotype expression and its modulation by chlorpyrifos: new insights into gene - toxic interactions". The thesis was directed by Dr. Maria Teresa Colomina and Dr. José Luís Domingo.

ABSTRACT

The extensive use and massive release of pesticides into the environment has led some authors wonder about the potential contribution of these substances on the emergence of neuropsychiatric and metabolic disorders. Currently, organophosphates (OP) – and chlorpyrifos (CPF) in particular - are the most widely used insecticides in the world. Given the heterogeneity of sources of exposure, most of the population is almost permanently exposed to CPF. Thus, a great body of epidemiological and experimental data have reported neurobehavioural deficits following exposure to the pesticide, and incipient evidence begins to suggest potential links between OPs and metabolic disorders. One of the critical issues is to establish whether there are subpopulations particularly susceptible to the detrimental effects of CPF. It has been proved that human apolipoprotein E (apoE) isoforms confer on their carriers varying vulnerabilities to metabolic diseases and neurodegeneration. The main objective of this thesis was to assess the behavioural and metabolic effects of both APOE genotype and CPF, as well as to determine whether the interaction between both factors contributed to these effects. To test this, adult mice expressing apoE2, apoE3 or apoE4 human isoforms were subchronically exposed to 2 mg/kg/day or 3.75 mg/kg/day CPF depending on the experimental phase. After exposure, animals were behaviourally tested, while the metabolic-disruptor role of CPF was also assessed in paralel. In general, APOE genotype influenced cognitive performance. The exposure to CPF altered metabolic functioning, and induced protracted attentional and motivational deficits. ApoE3 mice were the most vulnerable to the metabolic effects of CPF, and showed memory impairments after exposure. Furthermore, the pesticide reversed the lack of inhibitory control characteristic of apoE4 mice. Taken together, these results expand the existing literature on the behavioural and metabolic effects of CPF, and provide valuable information on gene – toxic interactions hitherto unknown.

The Director of TecnATox, Professor Jose L. Domingo, was included on the list (position #6) of the 30 most frequently cited European authors within the topic Toxicology as the magazine LabTimes compiled in its last update (#1, 2015). The list, which is referred to articles published between 2005 and 2011, is leaded by Prof. Mark T.D. Cronin, from the University of Liverpool (UK). England is the European country with more citations, while Spain occupies the 6th place. In fact, Prof. Domingo is the only Spanish researcher among the top-30 toxicologists across Europe. Link. Participation in a new H2020 project

TecnATox is one of the partners of the new Horizon2020 project EUROMIX, coordinated by the Dutch National Institute for Public Health and the Environment (RIVM). For the next 4 years, participants from 22 institutions and 5 associated partners, including the World Health Organization and the US Environmental Protection Agency, will be jointly evaluating the combined exposure to mixtures of multiple food‐related toxic substances and their combined risks, from different perspectives. The project is ultimately aimed at developing an experimentally verified, tiered strategy for the risk assessment of mixtures of multiple chemicals derived from multiple sources across different life stages.

Researchers from three different areas of TecnATox (Experimental toxicology, Human health risk assessment, and Food toxicology) will be fully involved in the development of the project, which includes PBPK modeling, in vivo experimental studies, validation with human data, total diet studies, and health risk assessment. In May 20-21, the kick-off meeting of EUROMIX took place at the Headquarters of RIVM (Utrecht, The Netherlands).

TecnATox hosted a keynote speech of Adrian Covaci

Prof. Adrian Covaci, the prestigious scientist of the University of Antwerp, Belgium, visited TecnATox facilities before the end of April. Taking advantage of his visit, Prof. Covaci gave the seminar entitled “In vitro metabolism of phosphate flame retardants and implications for biomonitoring” in April 30, 2015, 12 p.m., at Aula 113, ETSEQ.


Adrian Covaci is professor of Environmental Toxicology and Chemistry at the Toxicological Centre, University of Antwerp, Belgium. He is a very prolific researcher, having co-authored more than 350 papers in international scientific journals. He is Associate Editor of the journals Science of Total Environment and Food and Chemical Toxicology, as well as member of the Editorial Board of Environment International and Environmental Research. Keynote speaker at the ISHS Symposium

TecnATox was present at the III International Symposium on Organic Matter Management and Compost Use in Horticulture, which took place in April 20-24, in Murcia (Spain). The Symposium was organized by the International Society for Horticultural Science (ISHS) and the Group of Soil Enzimology and Biorremediation and Organic Wastes of the Spanish Council for Scientific Research (CSIC-CEBAS), being Dr. Miguel A. Sánchez-Monedero and Dr. Mariluz Cayuela the conveners.


Dr. Martí Nadal was invited to give a keynote lecture, entitled “Health risks associated to solid waste composting: From production to use in agriculture”. The speech was focused on 2 case-studies of health risks due to occupational exposure to compost, at different stages of its life cycle. Moreover, the poster “Airborne levels of VOCs and bioaerosols near a composting plant in Catalonia, Spain. Human health risk assessment” was also presented.

XXIII Scientific Course of Mediterrània-CIE

In April 15-17 2015, the 23rd edition of the Scientific Course, organized by Mediterrània-Centre d’Iniciatives Ecològiques and TecnATox, will be held in Tarragona. The conference, which is entitled “Toxicants: the dark side of the progress”, will gather together key speakers in the areas of environmental and food toxicology, cover a broad range of topics, from historical episodes of environmental pollution to the health impacts of food toxicants.

Partners of a new COST Action

TecnATox will be in the partnership of a new Trans-Domain COST Action, entitled “Network on technology-critical elements - from environmental processes to human health threats” (TD COST Action TD1407). The Action will create a network of scientists working and interested on technology-critical elements (TCEs), from an environmental perspective to potential human health threats, with the aim of defining the current state of knowledge and gaps, proposing priority research lines/activities, and acting as a platform for new collaborations and joint research projects.
COST is the longest-running European framework supporting trans-national cooperation among researchers, engineers and scholars across Europe. Participants from up to 16 European countries will take part in this specific COST Action, which will be coordinated by Dr. Antonio Cobelo (CSIC-Vigo). Dr. Martí Nadal will act as a member of the Management Committee.
